Mutation analysis of the K-RAS gene

Instructions for the patient

The most recent therapies for colorectal carcinomas use so-called “monoclonal” drugs, the efficacy of which is strongly influenced by the presence of mutations of the K-RAS oncogene: in particular, patients without mutations benefit from the therapy, whilst those with mutations do not generally respond. A test for the presence of K-RAS mutation is therefore necessary in order to decide whether a patient with a colorectal carcinoma is to be treated. This test can be carried out on the tissue samples used for the traditional histological examination.

Material to be provided

  • Inclusion in paraffin or unstained histological slides relating to the histological examination of the tumour. This material should be requested from the healthcare unit in which the diagnostic histological examination was carried out. Please ensure that the unit receives the attached form and returns it together with the material provided for analysis.
  • Copy of the diagnostic histological report.

If either of the above items is missing, the evaluation may be incomplete and in some cases the test may not be carried out.

Please note: for diagnostic histological examinations that have been carried out at the Campus Bio-Medico University Hospital, the request itself is sufficient and neither slides nor a copy of the report need be sent (Red order form).

Request for specialist SSN Services (The Italian National Health Service) (Red order form)

The form used to request the analysis must be filled out by an SSN physician with a copy filed in the relevant Outpatient Clinic’s Analysis Request Register. The request should include the words:

a.  “Estrazione di DNA o di RNA”
b.  “Conservazione di campioni di DNA o RNA”
c.  “Ricerca mutazioni gene K-RAS mediante sequenziamento (04 sequenze)”

In order for us to be able to keep in touch regarding the service requested, the referring physician contact telephone number should be included.

Delivery details

a)  The red SSN order form used to arrange a K-RAS gene mutation analysis should be delivered to the Outpatients Clinic reception desk (First Underground Level), from Monday to Friday, between 8 am and 10 am.
